Stachydrine Suppresses Viability & Migration of Astrocytoma Cells Via CXCR4/ERK & CXCR4/Akt Pathway Activity

Abstract: Stachydrine dose-dependently suppressed proliferation and colony formation in Res186 cells with no such effect on normal astrocytes. Stachydrine downregulated CXCR4 transcription through enhancing IκBα-based NF-κB inhibition. Stachydrine promoted apoptosis and cyclin D1/p27-associated G0/G1 phase arrest in a CXCR4/ERK- and CXCR4/Akt-dependent manner. Stachydrine suppressed MMP-associated migration and invasiveness via inhibiting CXCR4/Akt/MMP-9/2 and CXCR4/ERK/MMP-9/2 pathway activity. Stachydrine inhibits the… Show more

“…Indeed, adequate literature support exists on stachydrine, trigonelline, and the glucosinolate breakdown products. In vitro and in vivo studies show that stachydrine exhibits inhibition of the NF-κB signalling pathways [34,71,72]. Likewise, trigonelline showed antidiabetic effects and counteracted insulin resistance by suppressing inflammation [34,73].…”

“…Especially, Brassica vegetables are known for their preventive role against these inflammation related disorders, mainly due to their glucosinolate content. Glucosinolate hydrolysis products, the isothiocyanates, are known to play important roles in disease prevention by triggering antioxidant and anti-inflammatory responses, among others [31][32][33][34]. In our previous work, the nuclear factor (erythroid-derived 2)-like 2 (Nrf2) mediated antioxidant effect of M. subcordata different extracts and the presence, in these extracts, of phytochemicals such as glucosinolates was reported [35].…”

“…Stachydrine (STA), a bioactive constituent extracted from a medicinal herb Leonurus heterophyllus Sweet, has extensive pharmacological properties, including antioxidant, anti‐inflammatory, anticancer and cardioprotective properties . Recent studies have reported that STA inhibits NF‐κB and MAPK pathways in different cell types . As the NF‐κB and MAPK pathways are crucial for osteoclastogenesis and because STA exerts inhibitory effects on these pathways, we hypothesized that STA may represent a novel candidate for treating osteoclast‐related diseases by inducing the targeted suppression of osteoclastogenesis.…”

“…It was accounted for that stachydrine strongly affects provocative pathway [67]. Stachydrine stifles practicality and movement of astrocytoma cells by means of CXCR4/ERK and CXCR4/Akt pathway action [68]. Stachydrine ensures eNOS uncoupling and improves endothelial brokenness instigated by homocysteine [69].…”
