Stable expression of Leptospira interrogans antigens in auxotrophic Mycobacterium bovis BCG

Abstract: Mycobacterium bovis BCG has been proposed as an effective live vector for multivalent vaccines. The development of mycobacterial genetic systems to express foreign antigens and the adjuvanticity of BCG are the basis for the potential use of this attenuated mycobacterium as a recombinant vaccine vector. Stable plasmid vectors without antibiotic resistance markers are needed for heterologous antigen expression in BCG. Our group recently described the construction of a BCG expression system using auxotrophic comp… Show more

“…1). The most used auxotrophic targets for TB vaccination are the amino acids lysine (Nascimento et al 2009;Pavelka et al 2003), leucine (Borsuk et al 2007;Chambers et al 2000;Rizzi et al 2012;Seixas et al 2010), and methionine (McAdam et al 1995;Sampson et al 2011;Smith et al 2001), and the vitamin pantothenate (Chege et al 2013;Sambandamurthy et al 2002;Sampson et al 2011). Although widely used for studies in new TB vaccination strategies, auxotrophic approach has been used just once as an immunotherapy strategy for bladder cancer in an in vitro study (Begnini et al 2013).…”

“…The system described by Borsuk et al (2007) was further used for the expression of antigens from Leptospira interrogans [ 24 ] and M. bovis [ 25 ]. Seixas et al (2010) constructed two recombinant BCG Δ leuD strains, each expressing different leptospiral antigens.…”

“…Seixas et al (2010) constructed two recombinant BCG Δ leuD strains, each expressing different leptospiral antigens. Both strains were 100% stable in mice after 18 weeks of the experiment, whereas the BCG Pasteur strains harboring the same plasmids showed only a 7% stability in vivo in the same period post-inoculation [ 24 ]. A recombinant BCG strain overexpressing Ag85B was also developed using the Δ leuD auxotrophic system and was evaluated as a vaccine against bovine tuberculosis.…”

“…In addition, this method also allows for the development of auxotrophic strains, including strain, where genes involved in the synthesis of metabolites are deleted, strains without antibiotic resistance markers, and strains displaying stable expression both in vitro or in vivo (Borsuk et al 2007;Seixas et al 2010;Rizzi et al 2017).…”
