Stabilization of the gp120 V3 loop through hydrophobic interactions reduces the immunodominant V3-directed non-neutralizing response to HIV-1 envelope trimers

Abstract: To provide protective immunity against circulating primary HIV-1 strains, a vaccine most likely has to induce broadly neutralizing antibodies to the HIV-1 envelope glycoprotein (Env) spike. Recombinant Env trimers such as the prototype BG505 SOSIP.664 that closely mimic the native Env spike can induce autologous neutralizing antibodies (NAbs) against relatively resistant (tier 2) primary viruses. Ideally, Env immunogens should present broadly neutralizing antibody epitopes but limit the presentation of immunod… Show more

“…First, thermophilic proteins often have an increased number of hydrophobic residues at domain and oligomer interfaces, facilitating tighter packing of protein domains [46,47]. A similar strategy was applied to HIV-1 Env trimers to stabilize the trimer and prevent the exposure of non-NAbs [48][49][50][51][52][53]. For example, de Taeye et al avoided the spontaneous exposure of the V3 loop by increasing the hydrophobic interactions within the V3 domain and between the V3 and V1V2 domains, by Amino acid substitutions that help stabilize soluble native-like trimers.…”

“…Second, immunization with BG505 SOSIP.664 trimers induced a strong response against non-NAb V3 epitopes [10,50,53,70], leading to the hypothesis that this immunodominant V3-response interfered with the generation of bNAb responses. When rabbits were immunized with an improved version of the trimer, the BG505 SOSIP.…”

“…The Env V3 loop is a major target of non-neutralizing Abs, and is exposed in the Env open conformation. To reduce the V3 conformational dynamics, structure-based design was employed to strengthen hydrophobic packing, preventing V3 loop to adopt the open conformation [29,30]. Similarly, Kulp et al employed computational design to replace a network of buried hydrophilic residues by hydrophobic amino acids, thereby limiting V3 exposure [16 ].…”
