Stabilization of tetramethylbenzidine (TMB) reaction product at the electron microscopic level by ammonium molybdate

Abstract: The ability to use the tetramethylbenzidine (TMB) method for studying neuronal connections at the electron microscopic level is often difficult because the conditions of osmification and dehydration used in processing the tissue may result in significant loss and/or decreased electron density of the reaction product. In the present study, we report that stabilization of TMB reaction product with 5% ammonium molybdate (AM) prior to osmificating the tissue results in the formation of TMB-AM crystals that are man… Show more

“…Unfortunately, such sections are not very useful for examining m o r p h ological details. A n alternative is to use T M B to visualize the H R P , because the H R P / T M B reaction product is clearly visible as an electron-dense crystalline reaction p r o d u c t (Marfurt et al 1988).…”

“…Tissue sections were cut from blocks of visual cortex using a vibratome. These sections were then reacted with either diaminobenzidinc (DAB) (Graham and Karnovsky 1966) or with tetramethylbenzidine (TMB) (Marfurt et al 1988). The DAB reaction product was intensified with cobalt chloride and nickel ammonium sulfate (Adams 1981), after which all reacted tissue sections were osmicated and embedded in Araldite.…”

“…The sections then underwent a 20-minute reaction in the prereaction buffer with 0.3% H 2 O 2 added (Mesulam, 1978;Mesulam et al, 1980;Austyn and Gordon, 1981). This was followed by 15 minutes of incubation with 5% ammonium molybdate in ice-cold sodium acetate buffer to enhance the EM appearance of TMB crystals (Marfurt et al, 1988;Liang and Wan, 1989).…”

“…In rat 6, the segments were cut into 50-m-thick transverse sections on a Vibratome™ (Oxford Laboratories, Oxon, UK). These sections were processed for HRP histochemistry, using tetramethylbenzidine (TMB) as the chromogen, and treated according to a protocol for EM plastic embedding (Marfurt et al, 1988). The sections were inspected under the light microscope, and five sections from each segment were immediately transferred to 1% OsO 4 for 1 hr to stabilize the reaction product, dehydrated in alcohol, and embedded in Durcopan ACM (Fluka Chemie AG, Buchs, Switzerland).…”
