Stability of sheath blight resistance in transgenic ASD16 rice lines expressing a rice chi11 gene encoding chitinase

Abstract: Development of transgenic plants by introducing defense genes is one of the strategies to engineer disease resistance. Transgenic ASD16 rice plants harbouring rice chitinase chi11 gene, belonging to a PR-3 group of defense gene conferring sheath blight (Rhizoctonia solani Kuhn) resistance, were used in this study. Three T 2 homozygous lines (ASD16-4-1-1, 5-1-1, and 6-1-1) were identified from seven putative (T 0 ) transgenic lines expressing chi11 using Western blotting analysis. The inheritance of sheath blig… Show more

“…Moreover, the number of infection cushions formed on leaves was significantly lesser in transgenic lines expressing tlp and their F2 derivatives as compared to non-transgenic control. Similar kinds of results were obtained in detached leaf assays by earlier workers (Kumar et al 2003, Kalpana et al 2006, Maruthasalam et al 2007, Rajesh et al 2016.…”

“…In earlier reports, significantly lesser HRLH% and total lesion spread in transgenic lines expressing chi11 and/or tlp were reported (Kumar et al 2003, Kalpana et al 2006, Maruthasalam et al 2007. Recently, Rajesh et al (2016) has demonstrated the inheritance of sheath blight resistance (upto T4 generation) in transgenic ASD16 rice plants expressing a chitinase gene.…”

“…The control measures include the breeding of disease-resistant varieties, as well as the application of chemical, biological, or RNA pesticides. As China has no diseaseresistant rice varieties, chemical pesticides are mainly used to control rice sheath blight (Rajesh et al, 2016). However, due to the lack of disease-resistant resources of rice germplasm, Rhizoctonia solani has high genetic variability and wide host affinity, and can survive from one crop season to the next by forming dormant sclerotia, which increases the difficulty of controlling rice ShB.…”

“…alit has also been reported that there was a negative correlation between the mycelial growth of R. solani and chitinase activity in the antifungal assay conducted with recombinant LOC_Os11g47510, chitinase protein ( Richa et al, 2016 ). Very recently, Rajesh et al (2016) analyzed the sheath blight resistance in transgenic ASD16 rice lines expressing a rice chi11 gene encoding chitinase and found that transgenic line ASD16-4-1-1 gave a better protection against the sheath blight pathogen than the other two lines.…”
