Stability of Serum Eye Drops After Storage of 6 Months

Abstract: The concentration of a spectrum of cytokines involved in corneal epithelial wound healing and the epitheliothrophic effect of serum are not significantly changed after a prolonged storage of 6 months at -20 °C. Hence, it seems justifiable to provide patients with appropriate freezer capacity with a 6-month supply of autologous serum eye drops. Albumin--which is known to be relevant for ocular surface health--could serve as a cost-effective parameter for stability controls.

“…This decision is based on the significant effect that 50% AS has shown in several studies [1, 14, 50] and on the hypothesis that at higher concentrations some components can become harmful. In fact, the concentration of biologically active molecules is different in serum and tear fluids.…”

“…The amount of the substances in the eye drops should be maintained consistently during storage for the therapeutic effect of the ASEDs. Recently, a study reported that when unopened bottles were stored at −20°C, the concentrations of substances in ASEDs were not significantly changed for up to 6 months . In England and Wales, because ASEDs are made from the full donation (approx.…”

“…Recently, a study reported that when unopened bottles were stored at −20°C, the concentrations of substances in ASEDs were not significantly changed for up to 6 months. 34 In England and Wales, because ASEDs are made from the full donation (approx. 470 mL) of 1 blood unit, 16,17 a 5-or 6-month supply of ASEDs can be made, and a shelf life of 6 months has been adopted for unopened dropper bottles.…”

“…DMSO is a relatively stable protective agent to maintain the integrity of corneal cells, while sucrose molecules act as buffers in corneal protection, and ChS improves CEC activity in cryopreservation [98]. DMSO began to be treated as a tissue preservative to preserve cultured rabbit CECs by Smith [99]. Shortly thereafter, Mueller injected a preservation solution containing DMSO into the anterior chamber of an eyeball, placing the eyeball in a preservation solution containing glycerol.…”
