Stability of cytokines, chemokines and soluble activation markers in unprocessed blood stored under different conditions

Abstract: Background Biomarkers such as cytokines, chemokines, and soluble activation markers can be unstable when processing of blood is delayed. The stability of various biomarkers in serum and plasma was investigated when unprocessed blood samples were stored for up to 24 h at room and refrigerator temperature. Methods Blood was collected from 16 healthy volunteers. Unprocessed serum, EDTA and heparinized blood was stored at room (20–25 °C) and refrigerator temperature (4–8 °C) for 0.5, 2, 4, 6, 8, and 24 h after c… Show more

“…For instance, it was previously shown that the serum IL‐6 and TNF‐α half‐life was as short as 103 ± 27 minutes and 70 ± 11 minutes, respectively . Furthermore, it was also demonstrated that such half‐life varies according to time and temperature blood storage before sampling . Therefore, in future studies, sample collection should be performed in a standardized procedure in order to better define the potential clinical usefulness of some of those cytokines as inflammatory biomarker either at the time of diagnosis or in the disease course of BP.…”

“…Weaknesses of the study include suboptimal sample handling. In cord samples, marker concentrations could be influenced by a longer period before separation, and an increase in neopterin levels has been reported in similar conditions . However, because of our prospective design, any influence of suboptimal sample handling should not depend on outcome (T1D case status) and is therefore not likely to create any spurious association.…”

“…The transient nature of elevated circulating IL-21 might be due to two reasons: cytokines such as IL-21 are known to have short half-lives making its production and circulation a prompt event. 40,41 Secondly, IL-21 is transcriptionally regulated. For example, C-Rel-, a member of the NF Kappa B family, regulates the expression of IL-21 42,43 , suggesting that circulating IL-21 level may be tightly modulated.…”
