Stability Indicating Methods for Determination of Donepezil Hydrochloride According to ICH Guidelines

Abstract: It is a reversibly inhibitor of acetylcholinesterase, indicated for the treatment of mild to moderate dementia of Alzheimer's type 1. The structural formula of Donepezil Hydrochloride is as follows:A few methods have been reported for the determination of I including HPLC methods in tablets 2) and human plasma.3-7) Isocratic HPLC method with fluorescence detection at 390 nm with an excitation at 325 nm for determination of donepezil in plasma and microdialysate samples was developed. 3) Analysis and enantio… Show more

“…Based on the investigation of forced degradation behavior in our work, donepezil was found to be stable under mild acidic condition and alkaline condition, but sensitive to degradation with hydrogen per- oxide, which is also reported in earlier studies on stability of donepezil hydrochloride [17][18][19][20][21]. Even if it appears stable under mild conditions, in our experiment degradation was achieved in alkaline solution under more severe conditions, using high molarity alkaline solution and reflux.…”

“…The estimation of the limit of detection or quantitation was done on the basis of the ICH guidelines (Abbas, Fayez, & Abdel, 2006). The LOD (limit of detection, S/N ≥ 3) and the LOQ (limit of quantification, S/N ≥ 10) were measured according to the response and the slope.…”

“…Various strategies, such as HPLC‐UV [19], HPLC‐fluorescence [20] and HPLC‐MS detection [21], have been conducted for quantifying DPZ. Also, stability‐revealing techniques have been published [22].…”
