Stability-Indicating Determination of Tedizolid Phosphate in the Presence of its Active Form and Possible Degradants

Abstract: Tedizolid phosphate is an antibiotic prodrug that is metabolized into tedizolid which is used against various resistant bacterial strains. In this study, tedizolid phosphate was subjected to stress degradation conditions, namely, hydrolysis (neutral, acidic and alkaline), thermal, oxidative and photolytic ones. The prodrug was stable toward thermal and photolytic stress conditions, while it showed significant degradation upon applying oxidative and hydrolytic conditions. Two suggested chromatographic methods a… Show more

“…A reverse phase (RP) high-performance liquid chromatography (HPLC) with UV-detection (at 251 nm wavelength) was used for the quantification of TZP after slight modification of the reported HPLC method [ 46 ]. In brief, the HPLC system (Waters ® 1500-series controller, Milford, MA, USA) was used, which was equipped with a UV-detector (Waters ® 2489, dual absorbance detector, Milford, MA, USA), a binary pump (Waters ® 1525, Milford, MA, USA), automated sampling system (Waters ® 2707 Autosampler, Milford, MA, USA).…”

“…After 72 h, the orbital shaking was stopped and the vials were left for 24 h, then centrifuged at 6000 rpm for 20 min to settle down the undissolved solid remains [ 52 ]. Thereafter, supernatants were collected, filtered through 0.45 µ filtration unit (Millipore, Molsheim, France) and used to quantify the TZP contents by HPLC-UV method [ 46 ] as mentioned in the Supplementary Materials .…”

“…The electrode selectivity and specificity were tested by measuring the potential of 10 À 3 M TP and the same concentration of interfering ions [TDZ, commonly used additives and mentioned anions solutions by separate solution method (SSM)]. TDZ was prepared by refluxing an amount of TP in water for 35 h as reported in Moaaz et al [43]. The potentiometric selectivity coefficient (K Pot Primary ion, interferernt ) was used to measure the selectivity of the proposed electrodes.…”

“…This activity against different resistant bacteria requires drug monitoring in various processes as quality control analysis and wastewater assessment to limit future bacterial resistance. Three chromatographic methods have been reported for determination and separation of TP [41][42][43] and to the best of our knowledge, there has been no electrochemical method reported for its determination.…”
