Ssc-MiR-21-5p and Ssc-MiR-615 Regulates the Proliferation and Apoptosis of Leydig Cells by Targeting SOX5

Abstract: Leydig cells (LCs) are the predominant cells of androgen production, which plays key roles in spermatogenesis and maintaining male secondary sexual characteristics. Abnormal development of LCs affects androgen levels in vivo, affects fertility and may even lead to infertility. Little is known about the regulation mechanism on LCs’ development and maturation in domestic animals, especially the regulation of non-coding RNAs. In this study, we continued to dig deeper in the previous RNA-seq data of porcine LCs fr… Show more

“…Here, we identified miR-615-5p as a potential therapeutic approach to promote OPCs differentiation. Previous research has discovered that miR-615 can interact with several target genes, such as Sox5, IGF2, and EGFR, which play crucial roles in cell proliferation and differentiation [ 32 34 ]. Inhibiting miR-615 can release the negative regulation on these target genes, increasing expression levels and promoting cell growth.…”

“…Therefore, we established miRNA-centered lncRNA–miRNA, miRNA–mRNA, and circRNA–miRNA targeting relationship pairs and constructed corresponding networks based on these relationship pairs. In the lncRNA–miRNA–mRNA coexpression network, ssc-miR-615 is associated with cell proliferation and apoptosis ( Tang et al, 2022 ; Wu et al, 2020 ). ssc-miR-9820-5p can be adsorbed by circSLC41A1 in the form of sponge adsorption to promote SRSF1 and thus resist apoptosis of porcine granule cells ( Wang et al, 2022 ).…”

“…PTBP1 is expressed in almost all types of cells and belongs to the PTB family, including PTBP2 and PTBP3, as well as the subfamily of heteronuclear ribonucleoproteins (hnRNPs) as an RBP. Its gene is located on human chromosome 19p13.3 [ 41 , 42 , 43 , 44 ]. Structurally, PTBP1 is 531 amino acids long with an N-terminal nuclear shuttle domain and four repetitive RNA recognition motifs used to bind RNA and participate in the RNA-binding process [ 45 ].…”
