Sporulation at minimum specific growth rate inAspergillus nidulans chemostat culture predicted using protein synthesis efficiency estimations

Abstract: Ribosomal efficiency (RE) estimates provide a quantitative descriptor of intrinsic growth rate of cell populations using readily‐obtainable experimental data. In Aspergillus nidulans chemostat cultures, RE increased linearly with growth rate over the range 25—60% of maximum growth rate (μmax), consistent with increasing ribosomal usage with increased growth rate. Above 60%, RE did not in‐crease significantly, suggesting that all ribosomes were functional at 60% of μmax, further increases in growth rate, presum… Show more

“…A low growth rate is a common consequence of substrate-limited growth during prolonged fed-batch cultivation and poor mixing in large volumes of dense culture broth. Severe limitation of nutrients and low specific growth rates () trigger submerged conidiation and secondary metabolism in both fed and starved cultures of aspergilli (6,8,37,40). Retentostat cultures are continuous cultures with cell retention, which, instead of maintaining a fixed specific growth rate, forces into a transient decline to approach zero.…”

“…In brief, total RNA was extracted with perchloric acid according to the method of Herbert et al (18) and determined by the orcinol method (38) for pentoses, using yeast RNA as a standard. Biomass protein was extracted with hot alkali (8) and determined using the alkali-compatible DC Protein Assay (Bio-Rad) with bovine serum albumin (BSA) as a standard. Melanin was extracted with hot alkali and purified according to the method of de Cássia et al (12).…”

“…An increased q anabolism also implies an excess of ribosomal proteins at the 0.05 h Ϫ1 growth rate, as the amount of ribosomes for protein synthesis probably remains constant during the relatively short postpulse period. This excess capacity indicates a low ribosomal efficiency, which has been observed previously for low specific growth rates in both prokaryotes (17) and eukaryotes (6,35).…”

“…During subaerial growth, A. niger produces chains of black spores from biseriate conidiophores (55), but like other molds (15,16,56,62) it also conidiates in submerged culture in response to severe nitrogen or carbon limitation (15,27,46). More recent molecular studies have shown that the transcription factor BrlA plays a central role as a positive regulator of conidiation in liquid as well as aerial environments (1,2,25,40,67,74) and that nutrient starvation is associated with induction of brlA transcription (62).…”
