Spontaneous extraskeletal osteosarcoma with various histological growth patterns in the abdominal wall of an ICR mouse

Abstract: Extraskeletal osteosarcoma is extremely rare in mice. This case report demonstrates a spontaneous murine extraskeletal osteosarcoma that exhibited various histological growth patterns in an ICR mouse. At necropsy, the tumor mass was located in the abdominal wall and was 45 × 30 × 25 mm in size. Histopathologically, the tumor showed the following four growth patterns: a solid pattern of polygonal cells embedded in an osteoid eosinophilic matrix with calcification, an irregular sheet pattern of short spindle cel… Show more

“…An IHC stain for sp7, also known as Osterix, a transcription factor that induces mesenchymal cells to differentiate into osteoblasts and subsequently into osteocytes, is currently available. IHC evaluations of 2 extraskeletal osteosarcomas, 1 each in a mouse and a rat, found good positive staining of neoplastic cells (Ito et al 2016; Nagaike et al 2011). This suggests sp7 may be a useful marker in the diagnosis of highly cellular osteosarcomas in which osteoid formation is minimal or absent.…”

“…Osteosarcoma is a malignant tumor that normally develops in the bone tissue; however, there have been few reports on extraskeletal osteosarcoma (EO) in rats and mice 1 , 2 , 3 , 4 , 5 , 6 , 7 , 8 . Here, we describe the first reported case of spontaneous EO in the neck skeletal muscle of a mouse.…”

“…Osteocalcin and osterix are reliable markers for osteosarcoma, while myogenin is a marker for rhabdomyosarcoma. Some EO cases have been shown to be positive for vimentin, S-100, and α-SMA 2 , 3 , 4 , 5 , 6 , 7 . PCNA was used to determine proliferation activity.…”

“…Specifically, it has been reported in the skin (mouse) 1 , subcutaneous tissue (rat) 2 , 3 , thoracic cavity (rat) 4 , abdominal wall (mouse) 5 , stomach (rat) 6 , and cecum (rat) 7 . This paper describes the first case of spontaneous extraskeletal osteosarcoma detected in the duodenum of a mouse.…”

“…Osteocalcin and osteonectin are well-known reliable markers for osteogenic tumors, and some extraskeletal osteosarcomas are said to positively react with anti-vimentin, S-100 protein, and α-SMA antibodies 2 , 3 , 5 , 6 . Judging from the abovementioned results, the present tumor was diagnosed as an extraskeletal osteosarcoma originating from the duodenum.…”
