Hybrid seedlings from the cross Nicotiana tabacum × N. suaveolens, obtained by test-tube pollination and ovule culture, expressed lethality at 28°C. Characteristic lethal symptoms in these hybrid seedlings consisted of browning of hypocotyls and roots. One hundred and seventeen hybrid seedlings were eventually obtained by the use of test-tube pollination and ovule culture. Hybrid seedlings maintained at 36°C did not express any lethal symptoms. Hybrid seedlings used for further experiments were transferred to 36°C immediately after germination at 28°C. When hybrid seedlings cultured at 36°C were transferred to 28°C, their growth stopped and lethal symptoms were expressed. During the progressive expression of lethality, apoptotic features such as chromatin condensation, nuclear fragmentation and DNA fragmentation were detected. On the other hand, there was no sign of apoptotic cell death in the hybrid seedlings at 36°C. Based on the observation that the same lethal symptoms and the same apoptotic features were observed in the reciprocal cross, N. suaveolens × N. tabacum, we suggest that not only the underlying causes of hybrid lethality but also the underlying causes of apoptotic cell death are due to the interaction of coexisting heterogeneous genomes, rather than to the effect of cytoplasmic genes. Furthermore, the progression of apoptotic cell death in the cross N. tabacum × N. suaveolens began in stems and roots, followed by leaves.

We investigated the capability of Microcystis aeruginosa to cause apoptosis by pursuing morphological, molecular and physiological characteristics after exposure to H 2 O 2 . Microcystis proliferation was only weakly affected after exposure to 150 mM H 2 O 2 but cell numbers decreased dramatically after exposures of 250 and 325 mM H 2 O 2 . Cells exposed to 250 and 325 mM H 2 O 2 were examined using transmission electron microscopy, and they exhibited membrane deformation and partial disintegration of thylakoids. Correspondingly, fluorescence imaging of DNA by Hoechst 33342 staining revealed the condensation of nucleoid chromatin. Moreover, cellular injury was concomitant with dramatic decreases in photosynthetic efficiency (ratio of variable fluorescence to maximum fluorescence [Fv/Fm], maximum electron transport rate [ETRmax]) and elevated caspase-3-like activity after exposure of 250 and 325 mM H 2 O 2 . Terminal deoxynucleotidyl transferase Deoxyuridine 5-triphosphate nick end labelling (TUNEL) positive staining appeared in cells exposed to 250 mM and 325 mM H 2 O 2 , and the percentage staining increased with increasing H 2 O 2 concentration. These data suggested that M. aeruginosa exposed to H 2 O 2 underwent an apoptotic event. Additionally, cells exposed to H 2 O 2 had increased cytoplasmic vacuolation and nontypical DNA laddering. Increased caspase-3-like activity was not inhibited in the presence of the synthetic caspase inhibitor carbobenzoxy-valyl-alanyl-aspartyl-[O-methyl]-fluoromethylketone. Therefore, H 2 O 2 induced apoptotic-like cell death in a dose-dependent manner. Taken together, our results provided a novel mechanism for explaining cyanobacterial bloom dynamics in response to environmental stress. The results also contributed to the understanding of the origin and evolution of programmed cell death.

Hybrid seedlings from reciprocal interspecific crosses between Nicotiana nudicaulis and N. tabacum were not viable when cultured at 28°C; this an example of hybrid lethality. Characteristically, shoot apices and root tips of these hybrids began to brown immediately after germination at 28°C. However, hybrid seedlings did not exhibit any symptoms of lethality at 34°C, and their growth was normal at this higher temperature. When hybrid seedlings were cultured at 34°C and then transferred to 28°C, lethal symptoms emerged rapidly after the transfer. Therefore, hybrid lethality associated with reciprocal interspecific crosses between N. nudicaulis and N. tabacum was due to the interaction of coexisting heterologous genomes and not to a cytoplasmic effect. Furthermore, as the N. nudicaulis×N. tabacum seedlings die, features of apoptotic cell death, including chromatin condensation, nuclear fragmentation, and fragmentation of DNA, were evident. These observations indicated that hybrid lethality of N. nudicaulis×N. tabacum seedlings is accompanied by apoptotic cell death. This report is the first to demonstrate that hybrid lethality with apoptotic cell death results from N. nudicaulis×N. tabacum hybridization.
