BACKGROUNDCry1Ab has emerged as a bio‐insecticide to control Spodoptera litura (Lepidoptera: Noctuidae). However, the sublethal effects of Cry1Ab on the physiological changes and molecular level of S. litura have not been well documented. Our aims in this study were to assess the sublethal effect of Cry1Ab on S. litura, including midgut and Malpighian tubules as targets.RESULTSAfter sublethal Cry1Ab exposure, distinct histological alterations were mainly observed in the midgut. Furthermore, the results of comparative RNA sequencing and tandem mass tag‐based proteomics showed that, in the midgut, most differential expression genes (DEGs) were up‐regulated and significantly enriched in the serine protease activity pathway, and up‐regulated differential expression proteins (DEPs) were mainly associated with the oxidative phosphorylation pathway, whereas the down‐regulated involved in the ribosome pathways. In the Malpighian tubules, DEGs and DEPs were significantly enriched in the ribosome pathway. We proposed that ribosome may act as a universal target in energy metabolism with other pathways via the results of protein–protein interaction analysis. Further, by verification of the mRNA expression of some Cry protein receptor and detoxification genes after Cry1Ab treatment, it was suggested that the ribosomal proteins (RPs) possibly participate in influencing the Bt‐resistance of S. litura larvae under sublethal Cry1Ab exposure.CONCLUSIONUnder sublethal Cry1Ab exposure, the midgut of S. litura was damaged, and the proteotranscriptomic analysis elucidated that Cry1Ab disrupted the energy homeostasis of larvae. Furthermore, we emphasized the potential role of ribosomes in sublethal Cry1Ab exposure. © 2024 Society of Chemical Industry.

BACKGROUNDSpodoptera litura is one of the most harmful lepidoptera pests in China, which is difficult to control due to its strong resistance to the current frequently‐used insecticide species. Requirement of developing pesticides with novel toxicology mechanisms to control S. litura is urgent. The quassinoid of bruceine D display outstanding systemic properties and strong insecticidal activity against S. litura, which possess notable application potential for integrative management of S. litura. But the mechanism of toxicity remains unclear.RESULTSIn this study, we found that bruceine D exerts potent growth inhibitory activity against S. litura, disrupt the ecdysone and juvenile hormone titers, and causing long‐term adverse effects. Association analysis between transcriptomics and metabolomics suggested that bruceine D affected the digestion and absorption capacity of S. litura larvae by inducing a strong oxidative stress response and cell apoptosis in the intestine. Further analysis demonstrated that bruceine D can inhibit the activities of digestive and antioxidant enzymes and induce MDA and ROS overaccumulation in the midgut. Moreover, the protein level of Bax, cleavage caspase 3, and cytochrome c expressed in cytoplasm (cyto) were up‐regulated by bruceine D, while Bcl‐2 and cytochrome c expressed in mitochondria (mito) were down‐regulated. In addition, there was a noticeable increase in caspase‐3 protease activity. Histopathological observations revealed that bruceine D damages the structure of midgut epithelial cells and activates lysosomes, which subsequently disrupts the midgut tissue.CONCLUSIONOverall, our findings suggested that bruceine D induced excessive ROS accumulation in midgut epithelial cells. The resulting cell apoptosis disrupted midgut tissue, leading ultimately to reduced nutrient digestion and absorption in the midgut and the inhibition of larval growth.This article is protected by copyright. Chemical pesticides in crop protection pose long‐term threats to living beings due to their harmful side effects. Plant consortium, a mixture of diverse plant extracts and biomedicine, has gained attention for integrated pest management. Developmental formulations increase bioavailability of bioactive constituents, enhancing desired biological activities. The present study is undertaken to evaluate the pesticidal activity of the plant consortium composed of Zingiber officinale, Allium sativum, and Capsicum annuum against Spodoptera litura under laboratory condition. Followed by measuring pesticidal activity, ecotoxicity of prepared formulation was investigated. The plant consortium was prepared from the selected plant extracts through green science principles. The presence of possible bioactive compounds in plant consortium prepared from ethanolic extracts of selected plants was determined using Gas Chromatography Mass Spectrometry (GC‐MS). Pesticidal activity of the prepared consortium was investigated against S. litura by determination of cumulative mortality, lethal concentration. Active principles of pesticidal activity associated with plant consortium was readily extracted in ethanol. Prepared consortium revealed notable pesticidal activity against S. litura larval stages by recording high mortality and least LC50 and LC90. The plant consortium has not shown any ecotoxic effect which was confirmed by no sign of toxic effects on plant growth parameters of seedlings. Ecotoxicity of proposed consortium was also studied by measuring the soil parameters and phytotoxicity assessment. The present study suggests the possible utilization of presently proposed plant consortium as an effective pesticidal agent without affecting ecosystem.
