Splice Variant Specific Modulation of Ca V 1.2 Calcium Channel by Galectin-1 Regulates Arterial Constriction

Abstract: Rationale: Ca V 1.2 channels are essential for excitation-contraction coupling in the cardiovascular system, and alternative splicing optimizes its role. Galectin-1 (Gal-1) has been reported to regulate vascular smooth muscle cell (VSMC) function and play a role in pulmonary hypertension. We have identified Gal-1 multiple times in yeast 2-hybrid assays using the Ca V 1.2 I-II loop as bait. Objective: Our hypothesis is that Gal-1 interacts directly with Ca V 1.2 channel at the I-II loop to affect arterial constr… Show more

“…In the cardiovascular system, numerous alternatively spliced isoforms of L-type Ca 2+ channels occur, which leads to a non-homogeneous population of channels [16]. In addition, the channel population appears to be flexible and to adapt during different pathophysiological conditions including AHT and other [9,25,26,31,[37][38][39][40]. We hypothesize that changes in Ca v 1.2 channel populations is the molecular basis for the observations in this study.…”

“…Other experimental evidence suggests that downregulation of Ca V 1.2 channels affects the contractility, spreading, and migration of hVSMC via galectin-1, a protein implicated in regulating cell-cell and cellmatrix interactions. In particular, galectin-1 binds to a subpopulation of Ca V 1.2 channels that are devoid of alternatively spliced exon 9 [53] .…”
