Splenic immunoglobulin-secreting cells and their regulation in autoimmune mice.

Abstract: We have investigated in vitro the magnitude, nature, and regulation of spontaneous and mitogen-induced Ig secretion by splenic lymphocytes from several autoimmune murine strains (NZB, NZB X W, MRL/l BXSB) and appropriate, normal mice. All autoimmune strains had increased numbers of mature splenic B lymphocytes, which secreted and/or contained Ig, compared to age-matched normal strains. In NZB and NZB X W mice, the high frequency of mature B cells was apparent early in life, whereas in MRL/l and BXSB mice it wa… Show more

“…The absence of antibodies that react with BAFF-R-Fc would explain the different results with this form of BLyS blockade (14). Nonspecific immune stimuli induce increased autoantibody production in young (NZB ϫ NZW)F 1 mice (35). The development of antibodies to the therapeutic TACI could also occur in human lupus.…”

“…Both B and T cell abnormalities have been reported, and the relative importance of a given defect varies from strain to strain (1,2,5,6). For example, T helper cell hyperfunction plays a dominant role in the disease of MRL/1 mice (6-10), whereas primary B cell abnormalities have been identified in BXSB, NZB, and (NZB × NZW)F1 (NZB/W) mice (1,2,6,(11)(12)(13)(14). In fact, these latter mice, unlike MRL/1 mice (7,9), develop SLE despite neonatal thymectomy (1,2,7,15).…”

“…Though MRL-+/+ mice have much less severe autoimmune disease than do congenic MRLlprllpr (19,27,28), both mouse strains had similar levels of 8.4-kb MCF RNA. Thus, lpr did not increase 8.4-kb MCF expression on an autoimmune background.…”
