Spleen focus-forming Friend virus: identification of genomic RNA and its relationship to helper virus RNA

Abstract: The genome of the defective, murine spleen focus-forming Friend virus (SFFV) was identified as a 50S RNA complex consisting of 32S RNA monomers. Electrophoretic mobility and the molecular weights of unique RNase T1-resistant oligonucleotides (T1-oligonucleotides) indicated that the 32S RNA had a complexity of about 7.4 kilobases. Hybridization with DNA complementary to Friend murine leukemia virus (Fr-MLV) has distinguished two sets of nucleotide sequences in 32S SFFV RNA, 74% which were Fr-MLV related and 26%… Show more

“…(E) Schematic maps of the oligonucleotides homologous to the indicated clones of SFFV DNA are shown. The relative positions of the oligonucleotides on the genomic map of SFFV RNA were published previously (11). the 0.6-kbp LTR and 0.4 kbp from the 5' portion of the proviral SFFV genome, but contains only 1.4 kbp, rather than 2.0 kbp, of the env gene region from the 3' portion of the proviral genome.…”

“…When analyzed by the two-stage assay after cotransfection with amphotropic viral DNA, this subgenomic DNA fragment clone demonstrated full SFFV biological activity in the mouse (data not shown). The fragment begins on the left, with the area containing the SFFV-specific oligonucleotides 12, 19, and 6, which are related to the recombinant env sequences of mink cell focus-inducing viruses (12), and continues through the area of oligonucleotides 11, 8, 3, 21, 9, 10, and 7, which are more related to the parental ecotropic F-MuLV (11,12). Studies of nonglycosylated gp52 demonstrate an unglycosylated apoprotein of about 46,000 daltons (32).…”

“…The polytropic env gene probe was excised from a plasmid containing a 620-bp BamHI-to-EcoRI fragment cloned from a polycythemia-inducing strain of the Friend spleen focusforming virus (35). The defective spleen focus-forming virus corresponds to a deleted polytropic virus (24), and the BamHI-to-EcoRI fragment is homologous to an identically sized fragment in polytropic genomes (see Fig. 6).…”

“…The discrepancy in 1 2 the extent of homology could be explained by the sensitivity of the method; that is, differences of one or two nucleotides in RNA sequences would be recognized as different spots in fingerprinting analysis, but could be recognized as 1MW related sequences in hybridization studies. This explanation allows the possibility that some !I-~spots recognized as SFFV specific in this manu-34 script may be related, but not identical, to those of FN-2 RNA; this was shown to be the case in a recent paper by Evans et al (8).
