Spiroplasma membrane lipids

Abstract: Membranes of six spiroplasma strains belonging to different Spiroplasma species and subgroups were isolated by a combination of osmotic lysis and sonication in the presence of EDTA to block endogenous phospholipase activity. Analysis of membrane lipids showed that in addition to free and esterified cholesterol the spiroplasmas incorporated exogenous phospholipids from the growth medium. Sphingomyelin was preferentially incorporated from phosphatidylcholine-sphingomyelin vesicles or from the serum used to suppl… Show more

“…Hence, we made single (ScMreB5 I95A and ScMreB5 W96A ) and double (ScMreB5 I95A,W96A ) mutant constructs of these residues of ScMreB5 WT ( Fig. S3 ) and tested their binding using liposomes having lipid composition resembling the S. citri membrane ( Davis et al, 1985 ). Before the addition of liposomes, the protein samples were spun at 21,500 g to ensure that any protein aggregates were removed.…”

“…In order to decipher mechanistic details of ScMreB5-liposome interaction, we carried out a lipid specificity study. S. citri membrane consists of ~38% phosphatidylglycerol (DOPG, an anionic lipid) and ~14% phosphatidylcholine (DOPC, a neutral lipid) (35).…”

In comparison to ScMreB5 WT , the mutations did not abrogate liposome binding significantly.…”

“…Mcap cells are coccoid and their genome does not encode any fibril or MreB protein. In addition, Mcap and Spiroplasma membranes have a similar lipid composition (Davis et al, 1985;Rottem, 1980), an essential point when considering that cytoskeleton elements were found to be closely associated with the membrane (Trachtenberg et al, 2008).…”
