Spire localizationviazinc finger—containing domain is crucial for the asymmetric division of mouse oocyte

Abstract: Zinc plays an essential role in mammalian oocyte maturation, fertilization, and early embryogenesis, and depletion of zinc impairs cell cycle control, asymmetric division, and cytokinesis in oocyte. We report that zinc, via the actin nucleator Spire, acts as an essential regulator of the actin cytoskeleton remodeling during mouse oocyte maturation and fertilization. Depletion of zinc in the mouse oocyte impaired cortical and cytoplasmic actin formation. Spire is colocalized with zinc‐containing vesicles via it… Show more

“…The knockdown of AIP1 induced an increase in the amount of large actin‐positive patches around the spindle region. Similar phenotypes were observed in the depletion of actin nucleators, such as Formin2, 38 Spire 46 or treatment with CytoD, a widely used inhibitor of actin dynamics 38,40 . Moreover, in AIP1 knockdown cells, F‐actin accumulation colocalized with enriched total‐Cofilin in the shortened protrusions.…”

“…We found some reproduction-related genes in the GC group. For example, lhx9 encodes a key transcription factor in gonadal development ( Balasubramanian et al, 2014 ), and spir localization is critical for mouse oocyte asymmetric division ( Jo et al, 2019 ; Figure 1C , Figure 1—figure supplement 2E ). Hence, we annotated this group of cells as GCs.…”

“…For example, lhx9 has been shown as a key transcription factor in gonadal development(Balasubramanian, Bui, Xie, Deng, & Gan, 2014). spir localization is critical for mouse oocyte asymmetric division(Jo et al, 2019)( Figure 1C; figure supplement 2E ). Hence, we annotated this group of cells as germ-like cells.…”
