Sphingosine kinase 1 is up‐regulated in colon carcinogenesis

Abstract: Sphingosine kinase 1 (SK1) phosphorylates sphingosine to form sphingosine 1-phosphate (S1P), which has the ability to promote cell proliferation and survival and stimulate angiogenesis. The SK1/S1P pathway also plays a critical role in regulation of cyclooxygenase-2 (COX-2), a well-established pathogenic factor in colon carcinogenesis. Therefore, we examined the expression of SK1 and COX-2 in rat colon tumors induced by azoxymethane (AOM) and the relationship of these two proteins in normal and malignant intes… Show more

“…For example, clinical studies have documented expression of ABCC1 in a range of solid and hematological cancers and, in some cases, have correlated its expression with negative responses to treatment and poor disease outcome (36). SphK1 is also up-regulated in many types of cancer (37,38). Moreover, a monoclonal antibody that binds S1P with extremely high affinity and specificity significantly slows tumor progression and associated angiogenesis in several animal models of human cancer (39).…”

“…These molecules are expressed during tumor neovascularization and promote organization, survival or migration of endothelial cells (23)(24)(25)(26)(27)(28)(29)(30)(31). Other, IL-12-related proangiogenic molecules down-regulated in IL12rb2 ϩ vs. IL12rb2 Ϫ tumors were angiopoietin 1 and 2, eotaxin, procollagen type XVIII␣1, cox1, MMP9, and sphyngosin kinase 1 (32)(33)(34)(35)(36)(37)(38). Finally, IFN-␥ and epiregulin, a major regulator of vascular smooth muscle cell dedifferentiation (39,40), were found to be up-regulated 4.5-and 3.5-fold, respectively, in IL12rb2 ϩ vs. IL12rb2 Ϫ tumors grown in KO mice (Fig.…”

“…SphK1 mRNA content was found elevated in cancers, such as lung cancer, glioblastoma and colon carcinoma (Johnson et al, 2005;Van Brocklyn et al, 2005;Kawamori et al, 2006). On the contrary, down-regulation of SphK1 could arrest cell cycle and induce apoptosis (Pchejetski et al, 2005;Bonhoure et al, 2006;Cuvillier et al, 2007).…”
