“…Despite the correlation between age, sperm concentration and chromatin decondensation, DNA fragmentation or vacuoles in the sperm head were not related to age. The agerelated DNA fragmentation was found in several studies Gatimel et al, 2014;Liu et al, 2014); our data show an abnormal pattern as DNA fragmentation increased in men aged between 35 and 39 years and decreased in the older age men. We have no explanation for this finding, which may be a consequence of the effect of new lifestyle stressors, as previously mentioned.…”

“…The clinical significance of sperm morphology evaluation is, however, still a matter of debate. In fact, contrasting results have been reported by different laboratories worldwide (Menkveld, 2010(Menkveld, , 2013, mainly owing to different staining procedures (Gatimel et al, 2014;Henkel et al, 2008), implying that the simple examination with the optical microscope cannot provide accurate information on the morphological quality of the spermatozoon.…”

“…Although the origin of these vacuoles raises many questions, some strong correlations have been established between the morphology of the spermatozoon, in particular the presence of large vacuoles, and its nuclear quality (degree of chromatin condensation and/or DNA integrity, chromosomal content) [8]. However, the high frequency of sperm head vacuoles in fertile men highlights the physiological nature of some of these vacuoles [9,10]. The observation of sperm head vacuoles is used to select spermatozoa before microinjection: intracytoplasmic morphologically selected sperm injection (IMSI).…”

“…Gatimel et al did not find any significant differences between the vacuolar parameters and aniline blue staining, SCSA and TUNEL assay. In addition, they reported that all of the vacuole parameters values were identical in fertile and infertile men (Gatimel et al, 2014). There are limited studies about the relation between the presence of vacuole and sperm aneuploidy.…”
