Sperm-Hybrid Micromotor for Targeted Drug Delivery

Abstract: A sperm-driven micromotor is presented as a targeted drug delivery system, which is appealing to potentially treat diseases in the female reproductive tract. This system is demonstrated to be an efficient drug delivery vehicle by first loading a motile sperm cell with an anticancer drug (doxorubicin hydrochloride), guiding it magnetically, to an in vitro cultured tumor spheroid, and finally freeing the sperm cell to deliver the drug locally. The sperm release mechanism is designed to liberate the sperm when th… Show more

“…

Photodynamic therapy (PDT) functions when the light-excited photosensitizers transfer energy to oxygen molecules ( 3 O 2 ) to produce cytotoxic singlet oxygen ( 1 O 2 ) that can effectively kill cells or bacteria. [15] So far, researchers have also explored stimuli-responsive materials, [16][17][18][19] biomimetic materials, [20][21][22] and micro-organisms functionalized with nanomaterials [23] as MNMs to precisely transport cargos of drugs, [16,20,23,24] proteins, [18,22,25] and genes [19] in vitro and/or in vivo. Herein, an enzymatic micromotor based on hollow mesoporous SiO 2 (mSiO 2 ) microspheres is constructed as a mobile and highly efficient photosensitizer platform.

“…Magdanz et al [26] and Medina-Sanchez et al [25] reported the use of micromotors to guide or transport motile and immotile sperms to eventually overcome two of the main male infertility problems: oligozoospermia (low sperm count) and asthenozoospermia (poor sperm motility), respectively. In our group, a sperm-hybrid micromotor system was also developed as a new drug-delivery system to potentially treat diseases in the female reproductive tract ( Figure 3D & Table 2) [24,82]. In this work, DOX was loaded into bovine sperms by a co-incubation method.…”

“…Another strategy employed by some biohybrids and bioinspired microswimmers is the periodical deformation of their propulsion apparatus . This deformation leads to a change in the mobility matrix at each point in time.…”
