Sperm DNA fragmentation affects epigenetic feature in human male pronucleus

Abstract: To evaluate whether the sperm DNA fragmentation affects male pronucleus epigenetic factors, semen analysis was performed and DNA fragmentation was assessed by the method of sperm chromatin structure assay (SCSA). Human-mouse interspecies fertilisation was used to create human male pronucleus. Male pronucleus DNA methylation and H4K12 acetylation were evaluated by immunostaining. Results showed a significant positive correlation between the level of sperm DNA fragmentation and DNA methylation in male pronuclei.… Show more

“…When DNA fragmentation is more than 30%, there is an inverse relationship between DFI and level of H4K12ac in male pronuclei. These findings demonstrate that increased level of damage results in a decreased level of sperm H4K12 acetylation, because DNA strand breaks are likely to cause inappropriate localization of acetylated histones in male chromatin [192].…”

“…An elevated level of sperm DNA damage is a clinical issue in human reproductive medicine due to its associations with reduced fertilization rates [ 1 , 2 , 3 ], poor embryo development [ 2 , 4 , 5 , 6 , 7 , 8 , 9 ] and pregnancy loss [ 10 , 11 , 12 , 13 , 14 , 15 ]. Because sperm DNA damage may act as a significant barrier to successful conception, an effective treatment is wanting.…”
