Sperm 1: a POU-domain gene transiently expressed immediately before meiosis I in the male germ cell.

Abstract: Members of the POU-domain gene family encode for transcriptional regulatory molecules that are important for terminal differentiation of several organ systems, including anterior pituitary, sensory neurons, and B lymphocytes. We have identified a POU-domain factor, referred to as sperm 1 (Sprm-1). This factor is most related to the transactivator Oct-3/4, which is expressed in the early embryo, primordial germ cells, and the egg. However, in contrast with Oct-3/4, rat Sprm-1 is selectively expressed during a 3… Show more

“…We have previously identified an Oct-3͞4-like POU factor known as Sprm-1, which is specifically expressed in the developing male germ cell immediately before the first meiotic division (14). We show here that although Sprm-1 transcripts are detected before the meiotic reduction, the Sprm-1 protein is seen predominantly in the post-meiotic spermatid.…”

“…3E). Hlf3 (encoding histone H1t) is expressed in midto late-pachytene spermatocytes (22) and Sprm1 is transiently expressed from late-pachytene to diprotene (23). The transcripts from these genes were present in Cst Ϫ/Ϫ testes at the same levels as in the wild type.…”

“…The purified GST-GCM protein was then used in a binding site selection assay, in which the GST-GCM protein was incubated with a pool of oligonucleotides that exhibited randomized sequence at 8 bp in the center (15). GST-GCM bound to such an oligonucleotide (see Fig.…”

“…Protein-bound template was identified by elelctrophoretic mobility shift analysis and amplified by PCR in 34 cycles with 5Ј-GGATCCATTCCTAAG-3Ј and 5Ј-AGATCTGATCTGAGC-3Ј as primers. Throughout all six rounds of selection conditions were as previously described for the binding site selection of Sprm-1 (15).…”
