Spectroscopic Characterization of the NO Adduct of Hydroxylamine Oxidoreductase

Abstract: Hydroxylamine oxidoreductase (HAO) from the autotrophic nitrifying bacterium Nitrosomonas europaea catalyzes the oxidation of NH 2 OH to NO 2 -. The enzyme contains eight hemes per subunit which participate in catalysis and electron transport. NO is found to bind to the enzyme and inhibit electron flow to the acceptor protein, cytochrome c 554 . NO is found to oxidize either partially or fully reduced HAO, but NO will not reduce ferric HAO. Since NO can be reduced but not oxidized to product by HAO, NO is not … Show more

“…2d Inset: microwave frequency of 9.340 GHz, power at 2 W, receiver gain of 1 ϫ 10 5 , modulation frequency of 100 kHz, and modulation amplitude of 4 G. For both spectra of Fig. 2d, the signal for excess NO (g ϭ 1.98) has been subtracted (18). Spectra of cNP-NO, pH 5.6, and cNP-NO after first reducing the protein with sodium dithionite (pH 5.6), were prepared similarly to those at pH 7.5.…”

“…NOS is one of few heme-containing enzymes that make NO (11,12). NOS hydroxylates a guanidino nitrogen of Arg and then oxidizes the N -hydroxy-L-arginine intermediate (NOHA) to NO and L-citrulline (Scheme 1).…”

“…HAO was demonstrated to produce NO and N 2 O during the oxidation of NH 2 OH in vitro (11,12). More recently, HAO has been described to catalyze the reduction and oxidation of NO in vitro (3,9).…”
