Spectroscopic approach of the interaction study of ceftriaxone and human serum albumin

Abstract: Under physiological conditions, interaction between ceftriaxone and human serum albumin was investigated by using fluorescence spectroscopy and ultra violet (UV) absorption spectrum. From spectral analysis, ceftriaxone showed a strong ability to quench the intrinsic fluorescence of human serum albumin (HSA) through a static quenching procedure. The binding constant (k) is estimated as K=1.02× 10 3 M-1 at 298 K. Fourier transform infrared spectroscopy (FT-IR) spectroscopy with Fourier self-deconvolution techniq… Show more

“…At the same time, the hydrophobic cavity in site II is smaller than that in site I [ 40 ]. It has been shown in the literature that, in contrast to LV, which binds equally strongly in subdomains IIA and IIIA [ 41 ], CT, being a rather large molecule, binds much weaker in site II than in site I [ 42 , 43 ].…”

“…Absorption spectroscopy methods such as ultraviolet (UV) [ 14 , 86 , 87 , 88 , 89 , 90 ], infrared spectroscopy [ 14 , 88 , 90 , 91 , 92 ], spectrofluorimetry [ 14 , 93 ], microbiological methods [ 14 , 94 , 95 ], and capillary zone electrophoresis [ 96 ] have also been used for the analysis of ceftriaxone. When used as an identification technique, UV has limited selectivity because multiple compounds may have the same or similar spectra.…”

“…Cft-Na (Scheme 2, Supporting Information), a third-generation semisynthetic cephalosporin antibiotic, has broad-spectrum activity against Gram-positive and Gram-negative bacteria with low toxicity and long halflife. 12,13 Ceftriaxone is used for treatment of various infections caused by specific bacteria, such as blood poisoning meningitis and gonorrhea, 14 and infections caused by Neisseria gonorrhoeae and Salmonella typhi. 15 It is often used for the treatment of community-acquired or mild-to-moderate healthcare-associated pneumonia and is also a choice for treatment of bacterial meningitis.…”

“…Also, to cope with the drug solubility/permeability problem, the drug–bile salt combination may be helpful, so by extending our previous studies , with such systems herein, we have intended to study the interactional behavior of drug–bile salt system by taking ceftriaxone sodium (Cft-Na) as a model drug. Cft-Na (Scheme 2, Supporting Information), a third-generation semisynthetic cephalosporin antibiotic, has broad-spectrum activity against Gram-positive and Gram-negative bacteria with low toxicity and long half-life. , Ceftriaxone is used for treatment of various infections caused by specific bacteria, such as blood poisoning meningitis and gonorrhea, and infections caused by Neisseria gonorrhoeae and Salmonella typhi .
