Transdermal route has always been a route of choice for those drugs having limitations through oral route. Transdermal delivery of Ondansetron Hydrochloride (HCl) is one of the potential strategies, capable of maximizing the drug performance in patients with Hyperemesis Gravidarum facing issues with oral drug delivery. Current study focused on the development of matrix transdermal patches of Ondansetron HCl using solvent casting method with combination of different polymers and eugenol as penetration enhancer followed by physical characterization for various parameters such as weight variation, folding endurance, moisture content, moisture uptake, drug content, tensile strength, elongation break test, FT-IR studies, in-vitro percent drug release, stability studies. The result suggested that formulation F3 exhibited optimum results in respect to physical evaluation and in-vitro drug release exhibited release in concentration independent manner (62.2±1.3% upto 8 hrs). All formulations followed zero-order kinetics and Higuchi's model values showed 'diffusion' as predominant release mechanism. FT-IR analysis revealed that there are no potential chemical interactions between the drug and the polymers. Stability studies revealed promising result and the formulation (F3) was found to be stable at 40±2°C and 75±5% RH. The study indicated that transdermal delivery of Ondansetron HCl is a suitable way to increase therapeutic effectiveness.

Each method has its own demerit like derivative methods require long time for analysis; while HPLC method requires expensive columns, trained personnel for running analysis, using saline as an medium may incur some stability issues so a simple attempt was made for analysing such an effective drug using UV spectrophotometric method proves to be beneficial since the amount of solvent required will be less and very economical due to its ease in its operation, a part from that it is very least time consuming and worthful for analysing all the brands at a time in one single medium.…”

A simple, reliable, rapid and a novel method have been developed for determination of ondansetron hydrochloride. The method is truly based on measuring UV absorbance maximum of 210 nm using universal solvent i.e. double distilled water (DDW). Three different brands of ondansetron were purchased from market for the analysis. A standard solution of drug was initially prepared by dissolving 10 mg of standard drug in double distilled water to get concentration of 100μg/ml. The different aliquots of solution of three different brands were prepared in the range of 2-10 μg/ml. The percentage assay of each brand was calculated and their regression analysis was also performed. Among the three brands (Emeset, Zofer and ondem) Emeset showed high percentage assay value of 100.77 and remaining two brands showed same percentage assay value of 97.22. All the brands were found to be within the limit as per Indian pharmacopeia. The regression coefficient was with in tolerable limit i.e. 0.999.

Simple, sensitive and selective methods are developed for the spectrophotometric determination of drugs, viz., Montelukast sodium, Prasugrel, Ondensetron, Rosuvastatin calcium, Amlodepine besylate based on their reactivity towards N-bromosuccinimide (NBS). The method involves the addition of excess NBS of known concentration in the presence of 1M HCl, reactants are allowed to react and the unreacted NBS is estimated by the measurement in the decrease in the absorbance of the Rhodamine-B dye (λ max 557nm). This method has been applied for the determination of drugs in their pure form as well as in tablet formulations.
