Objective: Simple, sensitive, precise, reproducible and validated spectrophotometric methods have been developed for the quantification of montelukast sodium as leukotriene receptor antagonist drug, in pure and dosage forms (tablets). Methods: The methods use N-bromosuccinimide (NBS) as an oxidant and three dyes, amaranth, methylene blue, and indigo carmine, as auxiliary reagents. The three methods are based on oxidation reaction of montelukast sodium with a known excess of N-bromosuccinimide (NBS) in acid medium, followed by determination of unreacted NBS by the reaction with a fixed amount of dyes, amaranth, methylene blue, and indigo carmine followed by the measurement of the absorbance at 520, 664 and 610 nm, respectively. Results: Under the optimum conditions, linear relationships with good correlation coefficients (0.9993-0.9996) were found over the concentration ranges of 0.5-10, 1.0-12 and 0.5-8.0 µg/ml with a limit of detection (LOD) of 0.15, 0.3 and 0.14 µg/ml using amaranth, methylene blue, and indigo carmine methods, respectively. Intra-day and inter-day accuracy and precision of the methods have been evaluated. No interference was observed from the common tablet excipients. Conclusion: The proposed methods were validated in accordance with ICH guidelines and successfully applied to the analysis of montelukast sodium in dosage forms (tablets). The reliability of the methods was further ascertained by performing recovery studies using the standard addition method. Statistical comparison of the results obtained by applying the proposed methods with those of the reported method by applying student’s t-test and F-test revealed good agreement.

Aim:The aim of the study was to develop and validate a new simple, accurate, sensitive and specifi c bioanalytical spectrofluorimetric method for quantitative determination of donepezil hydrochloride (DH) in human plasma. Method: Estimation of DH in human plasma was done by spiked human plasma studies. Forestimation of DH in human plasma extraction was carried out using 5% TCA forprotein precipitation followed by liquid-liquid extraction with 5% IPA in n-Hexane.Under spectrofl uorimetric conditions delta value 65 applied in synchronous mode(medium sensitivity mode) and wavelength used in analysis was 389 nm. Result: Developed method found linear, in the concentration range of 50-105 ng/mL with correlation coeffi cient (R 2 ) 0.9965. Percentage recovery was found to be 75 %-85 %for DH. High recovery show that the method is free from the interference from plasma constituents. Conclusion: Proposed method can be used for estimation of DH in routine quality control laboratories for quantitative determination of DH in bulk as well as in human plasma. Further it can also be used to determine plasma DH concentration in drug monitoring or in pharmacokinetic investigations.

The current study reports about simple, rapid, sensitive and economic spectrophotometric method for determination of the antihypertensive, Carvedilol (CRV) and the antipsychotic, Aripiprazole (ARP) in pure form and pharmaceutical formulations. The suggested method is based on reaction of the studied drugs with haematoxylin reagent in the presence of boric acid to give a reddish violet chromogen at λmax= 560 nm. The percentage recoveries for the suggested method were 99.78±1.10 and 99.97±1.2 within the concentration ranges of 3-16 μg/mL and 8-32 μg/mL for Carvedilol and Aripiprazole respectively. Results show a minimum detection limit (LOD) of 0.63 and 1.79 μg/mL and a quantification limit (LOQ) of 1.91 and 5.42 μg/mL for Carvedilol and Aripiprazole respectively. The method showed excellent precision, with intra and inter-assay precisions of less than 2.0% at all concentrations. The suggested method was applied successfully for determination of the cited drugs in pure form and pharmaceutical formulations without interference of common pharmaceutical excipients with the method procedure. The suggested method was proved to be valid by analyzing both pure and pharmaceutical formulations, comparing the results obtained with those of the reported methods and demonstrating good accuracy and precision results.
