Spectrophotometric determination of polar and non-polar lipids in oleaginous yeast

Abstract: A spectrophotometric method of determining extractable lipids was compared with a gravimetric method to estimate intracellular polar and non-polar lipids in the oleaginous yeast, Candida curvata. The methods correlated well but the spectrophotometric method was simpler, required less sample, and its accuracy was not affected by non-lipid components.

“…The lipid content was determined as dry weight by Wang et al (1993). Two ml of 10% (v/v) perchloric acid (70%) was added to 50 mg of dried cells, the mixture was heated (100°C for 2 h), and the lipid was extracted by chloroform/methanol (2:1 v/v), the process was repeated for three times.…”

“…The spectrophotometric method has the additional advantage of being sensitive only to ester groups and hence non-lipid impurities are ignored. The results were obtained from spectroscopic method correlated well with those of the gravimetric method. (Wang L.W. et al, 1993) FUTURE PROSPECTS Biodiesel has become more attractive recently because of its environmental benefits, and the fact that it is made from renewable resources.…”
