Spectrophotometric assays for total antioxidant capacity (TAC) in dog serum: an update

Abstract: The aim of this review is to study the main spectrophotometric methods used to evaluate total antioxidant capacity (TAC) in serum samples of dogs. Total antioxidant capacity (TAC) is an analyte frequently used to assess the antioxidant status of biological samples and can evaluate the antioxidant response against the free radicals produced in a given disease. Trolox equivalent antioxidant capacity (TEAC), ferric reducing ability of plasma (FRAP), and cupric reducing antioxidant capacity (CUPRAC) are different … Show more

“…The volumes of the samples needed for TAC, GSH-Px and SOD determinations on the RXDaytona biochemical analyser were 100 μL of heparin plasma, 25 μL of whole blood and 30 μL of haemolysate, respectively. The TAS kit is based on the method of Miller et al [16]), which is one of the TEAC (trolox equivalent antioxidant capacity) ABTS (2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid)) based methods and has been previously used in dogs [18]. The results are expressed as mmol/L of Trolox (6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid) equivalents.…”

“…The given method shows the amount of low molecular weight antioxidants but does not include any compounds with thiol groups. 15 Based on Figure 3, ethanolic extracts had the highest reducing power followed by water. Autumn sample had the significantly highest reducing power in the range of 165.…”

“…Methods for determining the oxidative status include direct and indirect measurements of oxidants and antioxidants. Despite some disadvantages, measurements of TAC and TOS concentrations are a practical way to identify the oxidative status of an organism (13,32). One of the products formed during the oxidative damage-induced decomposition of polyunsaturated fatty acid is MDA (8).…”
