Specificity of insulin signalling in human skeletal muscle as revealed by small interfering RNA

Abstract: Insulin action on metabolically active tissues is a complex process involving positive and negative feedback regulation to control whole body glucose homeostasis. At the cellular level, glucose and lipid metabolism, as well as protein synthesis, are controlled through canonical insulin signalling cascades. The discovery of small interfering RNA (siRNA) allows for the molecular dissection of critical components of the regulation of metabolic and gene regulatory events in insulin-sensitive tissues. The applicati… Show more

“…In addition the silencing some protein molecules using the small interfering RNA technique (si-RNA) and the generation of multiple types of transgenic mice (MIRKO, FIRKO, LIRKO, NIRKO, BIRKO, VENIRKOcorresponding to the deletion of IR in muscle, adipocytes, liver, neurons, βcells or vascular endothelial cells), became a true industry in many countries (104). As expected, silencing or elimination of IRS-1 led to the generation of a true insulin-resistance, especially in conditions of high fat diet.…”

“…The study of cultured myocytes has provided important insights into mechanisms that regulate glucose uptake (1420). However, because cultured cells fail to replicate all properties of adult muscle, the ability to measure glucose uptake by single fibers from authentic muscle would prove valuable.…”

“…19 This occurs in spite of the insulin receptor sharing similar pathways with AT 1 and insulin growth factor-1 receptors with activation of mitogenic-activated-protein kinase, phosphatidyl inositol phosphoinositide 3-kinase/protein kinase B/Akt in the stimulation of cell growth and protein synthesis. 29,30 The mechanism(s) responsible for the tissue- and receptor- specific effect of the D 3 receptor on insulin receptor expression and function remains to be determined.…”
