Specific resistance upon lentiviral TRAIL transfer by intracellular retention of TRAIL receptors

Abstract: Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) selectively induces apoptosis in many transformed cells, suggesting TRAIL as an ideal candidate for cancer gene therapy. A main obstacle in cancer therapy is intrinsic or acquired therapy resistance of malignant cells. To study induction of resistance against TRAIL, we generated lentiviral vectors allowing efficient TRAIL expression and apoptosis induction in a variety of human cancer cell lines. Within days upon TRAIL overexpression, cells became… Show more

“…LV production was performed as previously described. 25,26 Transduction of MSC Cells were seeded in six-well plates at 2 Â 10 5 -2 Â 10 6 cells per well in 2-5 ml MSC medium containing 8 mg ml À1 polybrene and virus was added to the cells at different multiplicities of infection. The plates were centrifuged at 1200 r.p.m., 37 1C for 1 h and incubated at 37 1C.…”

“…In this case, TRAILsensitive cells would be killed by the TRAIL application, whereas the remaining resistant tumor cells would survive and proliferate. 91 Hypoxic environments, which are often present in growing solid tumors, are known to induce the up-regulation of hypoxia-inducible factor-1␣. This transcription factor binds to hypoxia-response elements, thereby inducing many hypoxia-response genes, 92 such as VEGF.…”

“…Many cancer cells are primarily resistant to apoptosis, which is usually due to down-regulation of the apoptosis pathways and/or activation of cell survival signals through genetic and epigenetic aberrations acquired during transformation (1,2). In addition, cancer cells acquire apoptosis resistance during chemotherapy (secondary or acquired apoptosis resistance) (3)(4)(5). Acquired apoptosis resistance has been an increasing concern because it has emerged as an important mechanism underlying acquired chemoresistance (6,7).…”

“…The molecular basis for acquired TRAIL resistance has not been well elucidated (4,5,18). We have recently established acquired TRAIL resistance in lung cancer cell lines by continuously exposing the TRAIL-sensitive lung cancer cells to nontoxic doses and gradually increasing the concentrations of TRAIL (19,20).…”
