Specific MR imaging of human lymphocytes by monoclonal antibody‐guided dextran‐magnetite particles

Abstract: Human lymphocytes were labeled with biotinylated anti-lymphocyte-directed monoclonal antibodies, to which streptavidin and subsequently biotinylated dextran-magnetite particles were coupled. This labeling resulted in a strong and selective negative contrast enhancement of lymphocyte suspensions at 2.0 T, caused predominantly by the specific increase of R2 with a small but significant specific increase of R1. The R1 was found to decrease with increasing field strength. The immunolabeling procedure described her… Show more

“…Monoclonal antibodies (mAbs) were the first targeting agents to exploit molecular recognition to deliver MNPs [125,126,127,128] and continue to be widely used due to their high specificity. Recently, the development of Herceptin ™ , an FDA-approved mAb to the HER2/neu (erbB2) receptor, has made it a popular targeting agent for nanoparticles [129,130].…”

“…MRI is an appealing noninvasive approach for early cancer diagnostics and therapeutics [10]. However, in clinical diagnosis, it is often the case that the diseased and healthy tissues are similar in composition and MRI technology alone will not always differentiate the two.…”

“…The strategy of targeting iron-oxide nanoparticles to selected receptors using mAb has been used to image lymphocytes in culture; a biotinylated anti-lymphocyte-directed mAb was conjugated to a biotinylated dextran-coated SPIO with a streptavidin linker, generating selective negative contrast enhancement of lymphocyte suspensions [21]. Kang et al [22] have imaged E-selectin expression in human endothelial cells by conjugating a F(ab′)2 fragment of the anti-human-E-selectin mAb to cross-linked iron oxide (CLIO) nanoparticles.…”
