Specific interactions of liposomes with PMN leukocytes upon incorporating tuftsin in their bilayers

Abstract: Incorporation of tuftsin derivatives, Thr‐Lys‐Pro‐Arg‐NH‐C18H37 or Thr‐Lys‐Pro‐Arg‐NH‐(CH2)2‐NH‐COC15H31, into an egg phosphatidylcholine/cholesterol liposome bilayer led to significantly enhanced binding of the liposomes to PMN leukocytes at 37°C but not at 0°C. Under identical conditions, no such enhanced binding of the liposomes was observed with erythrocytes and lymphocytes. These results demon‐strate that grafting of tuftsin on the liposome bilayer enables the liposome to recognize specifically the PMN le… Show more

“…The tube was dried to remove any trace of solvent. The lipid film was then hydrated and dispersed in a solution of appropriate quantity of p-tuftsin (7-8% by PC weight) in Tris-buffered saline (10 mM Tris containing 150 mM NaCl, pH 7.4) and processed by probe sonication (W-220) to get the desired preparation of liposomal tuftsin (Singhal et al 1984).…”

“…Egg phosphatidylcholine was isolated and purified by published procedures (18). Tuftsin modified by attachment of a palmitoyl residue to its C terminus through an ethylenediamine spacer arm (Thr-Lys-Pro-Arg-NH-(CH2)2-NH-CO-C15H31) was prepared as described previously (17).…”

“…The RFP A334 was found to remain unaffected by the presence of liposomes and detergent and was linear up to 100 ,ug of RFP. The amount of tuftsin incorporated in liposomes was estimated as described previously (17). The incorporation efficiency was found to be about 98%.…”

“…Structure-function studies of this tetrapeptide indicate that its binding and subsequent activation of the mononuclear phagocyte system (MPS) is dependent upon rather strict conservation of its molecular structure. Direct attachment of a fatty acyl group to the Arg residue, without any spacer arm, leads to modified tuftsin, which does not allow formation of liposomes, presumably due to perturbation of the phospholipid polar head group packing by the bulky Arg residue (Singhal et al 1984). However, the activity is largely retained if modifications are restricted only to the C-terminus (Gottlieb et al 1982).…”
