A multiplex PCR assay was developed to identify widespread poison-containing puffer fishes of the genus Lagocephalus: L. lunaris, L. spadiceus and L. inermis, using a novel KUGEN_ILSspec primer set which is specific for 12S ribosomal RNA (12S rRNA) and Cytochrome b (Cyt b) genes. KUGEN_ILSspec set is composed of fish positive-amplified primers that produced a 289-bp fragment while L. lunaris-, L. spadiceus-and L. inermis-specific primers produced 123-, 196-and 493-bp fragments, respectively. The sensitivity of this primer set was found to be high as it was capable of amplifying targeted DNA at concentrations as low as 1 ng/µL. Moreover, the primer set was shown to amplify intact DNA and species-specific fragments from heat-processed food. Consequently, multiplex PCR technique in combination with KUGEN_ILSspec primer set could offer an effective measure for detecting poisonous puffer fish contamination in both fresh and processed fish products, which will greatly aid in public health control and law enforcing endeavors.

Abstract. The objective of the present study was to examine the diversity and distribution of fishes in the Krueng Geumpang River.The survey was conducted from 1 st to 22 nd August 2015 with six sampling locations. Fish samples were caught using gillnets and fish traps. Data analyses performed in this study were the frequency of incidence (FOI), diveristy index (H'), and dominance index (C). A total of 88 individual fishes belong to 12 species and six familia. Tor soro is the dominant species in this river. There are two species of fish that widely distributed i.e. Tor soro (FOI = 66.7 %) and Neolissochilus thienemanni (FOI = 50.0 %). The Shannon-Wienner diversity index ranged from 0.00 to 2.05. The low value of the diversity index (H') can be caused by factors such as river morphology, poisoning, mining, and overfishing.

Biodiversitas 21: 1385-1390. Authentication of fishery products has relied mainly on DNA analysis of mitochondrial genes such as cytochrome c oxidase I (COI) DNA barcoding and cytochrome b (cyt b) gene fragments. The trend of sharks and rays food products trading in Indonesia significantly increased, due to their important role as protein source which may threaten the vulnerability status of some species. This study was aimed to determine the reliability of COI mini- DNA barcode and cyt b fragment for identification of shark species of heavily processed fishery products. We found the mini-DNA barcode was an effective tool to identify the shark species traded in local markets with most of the sample identifies as Carcharhinus falciformis. Our results highlight the urgency of applying DNA-based method as a routine method to control the labelling of Indonesian fish products and to manage sustainable fisheries.
