Species and tissue differences in serotonin glucuronidation

Abstract: 1. Serotonin is a UGT1A6 substrate that is mainly found in the extrahepatic tissues where some UGT1As are expressed. The aim of the present study was to characterize serotonin glucuronidation in various tissues of humans and rodents. 2. Serotonin glucuronidation in the human liver and kidney fitted to the Michaelis-Menten model, and the K values were similar to that of recombinant UGT1A6. However, serotonin glucuronidation in the human intestine fitted to the Hill equation, indicating that it is likely catalyz… Show more

“…Pooled rat microsomes from brain regions (n=8) and livers (n=3) were prepared as described previously. 21) β-Estradiol 3-Glucuronidation in Rat Brain Regions β-Estradiol 3-glucuronide levels were determined using a method described previously 22) with slight modifications. The incubation mixture contained 50 mM Tris (hydroxymethyl) aminomethane-HCl (pH 7.4), 5 mM MgCl 2 , 25 µg/mL alamethicin, 3 mM uridine 5′-diphosphoglucuronic acid, 5 µM β-estradiol, and microsomal proteins (brain: 0.5 mg/mL or liver: 0.01 mg/mL).…”

“…Bilirubin (5-50 µM), serotonin (1-10 mM), and APAP (1-10 mM) were glucuronidated mainly by Ugt1a1, 23) Ugt1a6, 21) and Ugt1a6 and Ugt1a7, 24) respectively, which were used as the Ugt inhibitors. β-Estradiol 3-glucuronidation was analyzed in the presence of each Ugt inhibitor.…”

“…The liver, small intestine, and kidney were removed from 8-week-old male Sprague-Dawley rats or C57BL/6N mice (Japan SLC, Hamamatsu, Japan). Pooled rat and mouse (n=5 each) liver, small intestine, and kidney microsomes were prepared as described previously, 12) and were stored at −80°C until used. Protein concentration was measured using the Lowry method with a Bio-Rad protein assay kit (Bio-Rad, Hercules, CA, U.S.A.).…”

“…Numerous studies have evaluated the in vivo effects of E2 using experimental animal models such as rats and mice, and many researchers have attempted to extrapolate the animal data to humans. [9][10][11] Since species differences in the kinetics of serotonin 12) and propofol glucuronidation 13) have been reported between human and rodents, it would be expedient to clarify whether similar differences occur in E17G formation.…”
