Speciation Analysis of Selenium Metabolites in Yeast-Based Food Supplements by ICPMS—Assisted Hydrophilic Interaction HPLC—Hybrid Linear Ion Trap/Orbitrap MSn

Abstract: A new method was proposed for speciation analysis for selenium metabolites in Se-rich yeast. The coupling of a normal bore (4.6 mm) hydrophilic interaction liquid chromatography (HILIC) column with a hybrid linear ion trap/orbital ion trap mass spectrometer allowed the detection of the selenium-isotopic pattern in mass spectra down to the intrascan abundance of 0.001 with the low- and sub-part per million (ppm) mass accuracy regardless of the concentration. The quantitative elution recovery was verified by onl… Show more

“…This technique has been used in an in vitro absorption, distribution, metabolism, and excretion workflow of cassette incubation for as many as 32 compounds, followed by quantitative bioanalysis using full scan acquisition (64). Targeting of compounds can also now be achieved on the fly for a variety of analyte groups, including those containing metals (76) or those with distinct isotopic patterns such as sulfur-and bromine-containing (77,78).…”

“…FTMS can reliably deliver mass accuracy below 1 ppm (54,(57)(58)(59)(60). This accomplishment can be aided by exploiting the use of certain ubiquitously present background ions.…”

“…Thus, HPLC-ICP-MS is practically useful for the screening and determination of column recovery and the monitoring of the purification efficiency of selenometabolites owing to its sensitivity and robustness. 38) As a complementary technique to HPLC-ICP-MS, electrospray ionization (ESI) and atmospheric pressure chemical ionization (APCI)-tandem mass spectrometry (MS-MS) have been used for the identification of unknown selenometabolites in yeast, [39][40][41] plant, 42) and animal samples. 43,44) The fact that Se produces a characteristic cluster of signals that reflect its isotopic pattern ( Se, 8.73%) has facilitated the scanning of Se-containing species by MS. As ESI is a softer technique than ICP, it can provide molecular information.…”
