Specialized Techniques for Site-Directed Mutagenesis in Cyanobacteria

Abstract: Synechococcus elongatus PCC 7942 is an excellent model system for studying the molecular mechanism of the circadian clock in cyanobacteria. The "plastic" genetic characteristics of this organism have facilitated the development of various methods for mutagenesis of its chromosome. These methods are based on homologous recombination between the chromosome and foreign DNA, introduced to the cyanobacteria by either transformation or conjugation. Here we describe different approaches to mutagenize the chromosome o… Show more

“…EMS mutagenesis of fresh ΔrpaA mutant cyanobacterial strains was carried out as in Kondo et al (66). Absorbance scans were taken of all cultures, as detailed in SI Materials and Methods, and genomic DNA was extracted by using standard methods (67). Before sequencing genomic DNA, the disruption of rpaA in all strains was verified by PCR (Fig.…”

“…Transformation and homologous recombination of vectors into S. elongatus were performed according to standard methods that take advantage of its natural competence (61)(62)(63). The genotypes of strains were confirmed by standard colony or whole cell PCR protocols using Taq DNA polymerase (NEB) or Phusion High-Fidelity DNA polymerase (NEB).…”

“…Null strains for cikA (AMC1426) or kaiC (AMC1511) were constructed by insertion of an inactivation cassette (42) or a Mu transposon (40). Cyanobacterial transformations were performed as described earlier (42). E. coli strains DH10B and BL21DE3 hosted plasmids, and they were grown as described previously (43).…”
