Special symposium: In vitro plant recalcitrance transcript profiling: A tool to assess the development of conifer embryos

Abstract: Embryogenesis is a phase of development that has been understudied at the molecular level, especially in conifers. Somatic embryogenesis, the asexual propagation of embryos, provides insights into development and has major applications in the forestry industry. Loblolly pine is the most important commercial species in the United States; however, it is recalcitrant to somatic embryogenesis methods. An appreciation of gene expression and the development of 'expression markers' would allow us to follow conifer em… Show more

“…This could partly explain a reduction in length of mature embryos. The suspensor is indeed suspected to play a role in the growth and nutrition of somatic embryos in Pinus taeda (Cairney et al 2000;Ciavatta et al 2001). Moreover, the age-related length reduction of CE we observed resembles to stress-induced reductions previously described.…”

“…These include not only traditional factors such as salt mixtures, auxins, and cytokinins, but also media supplementation with abscisic acid, brassinosteroids, ethylene inhibitors, specific sugar types, specific vitamins, organic acids, and gibberellin inhibitors (Pullman and Johnson 2002;Pullman et al 2003bPullman et al , c, 2005bPullman et al , c, 2006Pullman et al , 2008Gupta et al 2004). Control of environmental factors including, water potential, pH, adsorption of medium components by activated carbon (AC), pressurized and/or gas-tight enclosure, absence of gelling agent (liquid media), and study of embryo gene expression patterns, have led to further initiation improvements (Xu et al 1997;Cairney et al 2000;Pullman and Johnson 2002;Peter 2002, 2006;Pullman et al 2005b;Pullman and Skryabina 2007;Cairney and Pullman 2007). Employing combinations of both media and environmental agents has permitted great increases in embryogenic tissue initiation.…”
