Spatial contextual recognition memory updating is modulated by dopamine release in the dorsal hippocampus from the locus coeruleus

Abstract: Detecting novelty is critical to consolidate declarative memories, such as spatial contextual recognition memory. It has been shown that stored memories, when retrieved, are susceptible to modification, incorporating new information through an updating process. Catecholamine release in the hippocampal CA1 region consolidates an object location memory (OLM). This work hypothesized that spatial contextual memory updating could be changed by decreasing catecholamine release in the hippocampal CA1 terminals from t… Show more

“…3E, F). Given that NA and DA both perform critical roles in dorsal hippocampal neuronal activity, plasticity, and memory process, 60,61,63,64,85–87 our results suggest that the dorsal hippocampal NA and DA release contributes to the light-exercise-induced hippocampal activation and beneficial effects, such as hippocampal neurogenesis and spatial memory performance. 9,29,30 Notably, NA and DA have been recognized as important factors in memory consolidation, 63,86 as shown by the synaptic tagging and capture hypothesis, 88,89 raising a hypothesis that a single-bout of light-intensity running could facilitate memory retention through noradrenergic and dopaminergic dorsal hippocampal activation.…”

“…6366,85,92 On the other hand, VTA TH neurons have sparse input to the dorsal hippocampus and do not have a significant impact on spatial memory. 63,64 However, hippocampal DA from VTA TH neurons is still reported to be involved in hippocampus-related functions, such as spatial memory and aversive memory. 70,71,93 The significance of LC TH and VTA TH neurons for the dorsal hippocampal functions remains elusive; thus, future studies using viral-tracing techniques and chemogenetic/optogenetic manipulation are needed to investigate the noradrenergic and dopaminergic mechanisms underlying the beneficial effects of light exercise.…”

“…Recently, we demonstrated that optogenetic inhibition of catecholaminergic projections arriving at the dorsal CA1 hippocampus, coming from the locus coeruleus but not from the ventral tegmental area, impedes object location memory updating. Significantly, the pharmacological blockade of hippocampal β-adrenergic receptors with propranolol hinders memory expression without altering memory updating, whereas D1/D5 receptors blockade, by SCH 23390 administration, impairs memory expression and updating ( Gálvez-Márquez et al, 2022 ). These results suggest that dopaminergic activity arising from the locus coeruleus modulates both memory expression and updating when new contextual information is presented.…”

“…8 The signal transmission of hippocampal dopamine is deeply involved in spatial cognitive processes, 9,10 and the updating of spatial contextual memory in the posterior hippocampus is also modulated by dopamine originating from the locus coeruleus. 11 Existing research has revealed that information related to rewards triggers dopamine release, thereby influencing hippocampal representations. 12 However, the role of dopamine extends far beyond this.…”

“…Dopaminergic neurons from the midbrain project direct synaptic projections to the hippocampal region. , Dopamine regulates hippocampal function and serves as a critical precursor to long-term potentiation, playing a vital role in signaling motivational events and aiding the hippocampus in forming long-term memories .
