Spatial, a gene expressed in thymic stromal cells, depends on three-dimensional thymus organization for its expression

Abstract: Although the importance of thymic stroma in thymopoiesis has been recognized, the underlying molecular details regarding stromal cell biology remain obscure. To study this area, we have cloned genes expressed in thymic stromal cells. Spatial is alternatively spliced to generate two mRNAs in thymus and lymph node (LN) but it is not expressed in the spleen. In mouse embryos, the short form begins expression at day 10 while the long form is not detected until day 12. Both mRNAs encode proline rich proteins and th… Show more

“…2C). Since computational analyses predict an homology to homeobox and POU domain transcription factors [13], we first suggested a nuclear function of Spatial specific to mature mTEC. In order to see if all mature mTEC are also Spatial positive, we quantified on WT adult thymic sections the respective cellularity of Aire 1 and Spatial 1 cells presents in medullary areas, detected with K5 or MTS10 markers (Fig.…”

“…In the thymus, Spatial expression was first documented by Flomerfelt et al [13]. Accordingly, it has been described that in mouse embryos, Spatial-g expression begins at day 10 while Spatial-a is not detected until day 12.…”

“…This analysis led to the identification of three adult variants of the Spatial gene expressed in the thymus (GenBank accession nos. AF257502, AF257503 and AY243457) [2,3]. In addition, we also described two other isoforms isolated in the testis (GenBank accession no.…”
