SPARCL1 Influences Bovine Skeletal Muscle-Derived Satellite Cell Migration and Differentiation through an ITGB1-Mediated Signaling Pathway

Abstract: As an extracellular matrix protein, secreted protein acidic and rich in cysteine (SPARC)-like 1 (SPARCL1) is involved in various cell functions. It was previously implicated in bovine skeletal muscle-derived satellite cell (MDSC) differentiation; however, the underlying mechanism remains unknown. In this study, immunoprecipitation and mass spectrometry revealed that integrin β1 (ITGB1) combines with SPARCL1. Further, co-immunoprecipitation demonstrated that SPARCL1 interacts with ITGB1. Cell scratch assays exp… Show more

“…6e). Besides, the SCs were enriched for genes associated with SC differentiation, regeneration and survival (SPARCL1, CRYAB, and GPX3) [39][40][41] , as well as genes involved in cell-cycle inhibition (NUPR1, C10orf10/DEPP) 42,43 to maintain their quiescence (Fig. 6e).…”

“…Another important linkage involves the SPARC proteins (secreted protein acidic and rich in cysteine, osteonectin), extracellular matrix proteins that are regulatory but do not contribute significantly to ECM structure [188]. SPARC-like protein 1 (SPARCL-1) is secreted by human SCs [164] and has been shown to promote SC activation via focal adhesions.…”

“…Paxillin is a major component of FA and is responsible for mediating the transduction of extracellular signals to intracellular signals, a process triggered by the interaction of the ECM with integrins [ 8 ]. One recent study using a bovine model found that SPARCL1, a component of ECM2, binds to integrin β1 via mediated overexpression of phosphorylated FAK and paxillin to promote skeletal muscle satellite cell migration [ 9 ].…”
