Sp1- and Sp3-mediated Transcriptional Regulation of the Fibroblast Growth Factor Receptor 1 Gene in Chicken Skeletal Muscle Cells

Abstract: Expression of the fibroblast growth factor receptor 1 (FGFR1) gene in skeletal muscle is positively regulated in proliferating myoblasts and declines during differentiation. We have characterized the cis-regulatory elements in the proximal region of the FGFR1 promoter which render positive transcriptional activity. Multiple elements between ؊69 and ؊14 activate the FGFR1 promoter. Myoblast transfections revealed that potential Sp transcription factor binding sites are required for promoter activity. Electromob… Show more

“…Blots were washed as before and developed by chemiluminescence (Pierce). For western blots of NFATc1 and MEF2A, nuclear protein extracts were collected from PM and MA muscle fiber cultures as previously described (Parakati and DiMario, 2002). Protein concentrations were determined using a BCA protein assay (Pierce).…”

“…The NFAT oligonucleotide consisted of the sequence 5′ GAGGCAGAAA-GGAAAGCTCTCAGT 3′ and its complement. EMSA reactions were prepared as previously described (Parakati and DiMario, 2002). MEF2A and NFATc1 antibodies (4 µg) were added to the reaction mixtures and incubated overnight at 4°C.…”

“…Sp1 is broadly expressed and activates a wide variety of constitutively expressed and differentially regulated genes. For example, Sp1 activates the avian FGFR1 promoter in proliferating myoblasts (18). However, other Sp and KLF proteins (e.g.…”
