SOX2 has a crucial role in the lineage determination and proliferation of mesenchymal stem cells through Dickkopf-1 and c-MYC

Abstract: SOX2 is a well-known core transcription factor in embryonic stem cells (ESCs) and has an important role in the maintenance of pluripotency. Recently, SOX2 expression has also been reported in adult stem cells (ASCs), but the role of SOX2 in ASCs remains unknown. In this study, we examined the molecular mechanisms of SOX2 in human mesenchymal stem cells (hMSCs), a type of ASCs, by performing inhibition studies. SOX2 inhibition resulted in altered cell growth and differentiation capabilities. These changes coinc… Show more

“…[42][43][44] let-7 targets LIN28, HMGA2, and MYC, which are important for maintenance of pluripotency, to promote stem cell differentiation. 40,[45][46][47][48] Recently, Wei and colleagues showed that let-7 enhances osteogenesis and bone formation of human MSCs by regulating HMGA2, suggesting that let7a suppresses these genes to regulate the functions of MSCs. In this study, we observed that let-7a inhibited the proliferation and self-renew of MSCs, whereas promoted the differentiation of MSCs.…”

“…Pluripotent transcription factors, SOX2, OCT4, and NANOG, key regulators in ESCs, maintain self-renewal and multipotency of adult stem cells [27][28][29][30]. Specifically, maintenance of self-renewal and multipotency of hMSCs requires Sox2 [31,32] through mechanisms involving sirtuin [18] and Wnt [31] signaling. Wnt signaling can cross talk with the antiapoptotic/prosurvival pathway, PI3k/Akt, promoting self-renewal and expansion of stem cells [33].…”
