Sox2 and Pou2f1 interact to control lens and olfactory placode development

Abstract: Sox2, which encodes an SRY-like HMG box transcription factor, is critical for vertebrate development. Sox2 mediates its transcriptional effects through the formation of complexes with specific co-factors, many of which are unknown. In this report, we identify Oct-1, encoded by the Pou2f1 gene, as a co-factor for Sox2 in the context of mouse lens and nasal placode induction. Oct-1, Sox2, and Pax6 are co-expressed during lens and nasal placode induction and during subsequent developmental stages. Genetic combina… Show more

“…At E10.5, Foxg1 is expressed throughout the invaginating olfactory placode in a pattern similar to a putative stem cell marker, Sox2 (Fig. 1 A) (Donner et al, 2007). We next localized Foxg1 protein by immunohistochemistry, which revealed two important findings.…”

“…There are examples that these factors interact to coregulate genes outside ES cells. POU2F1 and SOX2 have been shown to function synergistically in the regulation of the PAX6 and nestin genes later in neural development (Donner et al 2007;Jin et al 2009). SOX2 is widely expressed and therefore present with POU2F1 in many more tissues than POU5F1, including cell types and cell lines (e.g., HeLa) where POU2F1 is the only POU factor detectable (M Gemberling and L Ferraris, unpubl.).…”

“…The ectodermal enhancer (EE) was shown to direct Pax6 expression in the surface ectoderm, lens and cornea (Kammandel et al, 1999;Williams et al, 1998). The EE contains binding sites for the Meis (Zhang et al, 2002), Sox (Aota et al, 2003;Donner et al, 2006a), Six3 and Oct family of proteins (Donner et al, 2006a), as well as a single Pax6-binding site (Aota et al, 2003). Transgenic expression supported by this enhancer is initiated in the surface ectoderm at E8.75 in mouse, i.e.…”
