Southern blot analysis of T‐cell receptor gene rearrangements in cynomolgus monkeys, and identification of a progenitor cell HPRT mutation

Abstract: Increases in peripheral blood T-lymphocyte HPRT mutant frequency may reflect either a number of independent HPRT gene mutational events or clonal proliferation of a single HPRT mutant. Sequence analysis of HPRT mutations in conjunction with T-cell receptor (TCR) gene rearrangement pattern analysis can distinguish these possibilities. Our laboratory previously characterized a nonhuman primate model for in vivo mutation studies using the clonal HPRT mutation assay. In the present study we report the use of probe… Show more

“…For example, animal 7008 had a mean spontaneous MF of 16.4 ϫ 10 -6 , much higher than the historical spontaneous MF in our studies (mean MF Ϯ SD ϭ 2.9 Ϯ 2.9 ϫ 10 -6 based on 131 determinations in 33 animals [Zimmer and Aaron, 1997]). It was ultimately determined that the high MF in this animal was due to a proliferating prethymic mutant clone [Mattano et al, 1995]. Both 9179 and 9180 exhibited low CE throughout the study both before and after treatment, making it difficult to obtain sufficient numbers of mutants.…”
