Sortilin, Encoded by the Cardiovascular Risk Gene SORT1, and Its Suggested Functions in Cardiovascular Disease

Abstract: Several genome-wide association studies have linked novel loci to a wide range of cardiovascular phenotypes including low-density lipoprotein (LDL)-cholesterol, early onset myocardial infarction, coronary artery calcification, coronary artery stenosis, and abdominal aorta aneurysm. Especially, one locus, namely, 1p13.3, has attracted much attention. This locus harbors four candidate genes, CELSR2, PSRC1, MYBPHL, and SORT1. SORT1 encodes sortilin, a type I sorting receptor that has recently been implicated in L… Show more

“…Sortilin 1 (SORT1) encoding the transmembrane type I receptor sortilin is a risk gene for hypercholesterolemia and myocardial infarction [7, 87, 90]. Sortilin is highly expressed in a number of tissues in a distinct and dynamic fashion [17].…”

“…Plasma CELSR2, PSRC1, and MYBPHL, and a subsequent study has shown that SORT1 affects lipid metabolism 11) . SORT1 encodes sortilin, a type 1 transmembrane protein 12,13) , which acts as a multiligand receptor and is linked to systemic lipoprotein metabolism and PCSK9 secretion [14][15][16][17] . Lowering LDL cholesterol levels is currently considered the most effective strategy for preventing CAD.…”

“…The role of sortilin in VLDL-B100 secretion is controversial [1], and inconsistencies have been summarized [2, 3]. Some studies suggest sortilin serves as a chaperone where sortilin is hypothesized to facilitate secretion of VLDL-B100 [4].…”
