Sonic hedgehog signaling in the development of the mouse hypothalamus

Abstract: The expression pattern of Sonic Hedgehog (Shh) in the developing hypothalamus changes over time. Shh is initially expressed in the prechordal mesoderm and later in the hypothalamic neuroepithelium—first medially, and then in two off-medial domains. This dynamic expression suggests that Shh might regulate several aspects of hypothalamic development. To gain insight into them, lineage tracing, (conditional) gene inactivation in mouse, in ovo loss- and gain-of-function approaches in chick and analysis of Shh expr… Show more

“…Generation of JBST neurons from iPSCs. We attempted to generate ARH-like neurons from the JBST iPSCs but were unable to generate NKX2.1-positive neuronal progenitors from which POMC and NPY neurons derive (38,39), further suggesting the importance of RPGRIP1L in the development of ARH neurons. Subsequently, we utilized a dual-SMAD inhibition protocol (40) to direct differentiation of the JBST iPSCs into neurons positive for neuromarkers Nestin, neuron-specific class III β-tubulin (TUJ1), and Neurofilament (NF) (Supplemental Figure 3C).…”

“…Shh, which is secreted by prechordal mesoderm at E7.5 and by hypothalamic neuroepithelium at E8.5, plays a critical role in the development of the ventral portion of the neural tube (10). Shh signaling increases Nkx2-1 expression (11), which, as mentioned above, plays a critical role in the development of the neural tube ventral midline (9), and deletion of Shh in mice with the Nkx2-1-Cre driver (12) affects the development of POMC neurons.…”

“…Transcription factors expressed in the ventral forebrain, like Nkx2.1 and Six3, are then necessary to establish the hypothalamic character of the region 3,4 . Shh produced by the ventral forebrain is also necessary to stimulate proliferation and neuronal differentiation in the future hypothalamus 5,6,7,8 .…”
