Sonic hedgehog signaling in kidney fibrosis: a master communicator

Abstract: The hedgehog signaling cascade is an evolutionarily conserved pathway that regulates multiple aspects of embryonic development and plays a decisive role in tissue homeostasis. As the best studied member of three hedgehog ligands, sonic hedgehog (Shh) is known to be associated with kidney development and tissue repair after various insults. Recent studies uncover an intrinsic link between dysregulated Shh signaling and renal fibrogenesis. In various types of chronic kidney disease (CKD), Shh is upregulated spec… Show more

“…To explore how fibroblasts become activated rapidly after injury, we sought to identify the trigger responsible for fibroblast activation after IRI. To this end, we studied the potential involvement of Shh because earlier studies implicate it as a tubule‐derived growth factor for interstitial fibroblasts in chronic kidney disease (CKD) (25, 33, 34). As shown in Fig.…”

“…Fig 1a–b). Hh ligands, including Sonic hedgehog (Shh) and Indian hedgehog (Ihh), signal by binding to their transmembrane receptor patched (Ptch1) [13], which activates intracellular transcription factors called Gli proteins [14]. These transcription factors include Gli1, Gli2 and Gli3.…”

“…Cell-to-cell communication is well known to play a crucial role in guiding the proper development of multicellular organisms during the embryonic stage and in the maintenance of tissue homeostasis in adults under normal physiologic conditions. [12][13][14][15] Such a communication is often achieved through the engagement between secreted soluble cues from a particular cell and the neighboring responding cells via both autocrine and paracrine fashions. Earlier studies have shown that sonic hedgehog (Shh) and Wnt ligands are upregulated in diseased kidneys and could play a role in mediating fibroblast activation in the pathogenesis of CKD.…”
