An efficient protocol for plant regeneration through somatic embryogenesis was established from in vivo leaf explants of Swertia chirayita, a critically endangered medicinal herb. The highest frequency (76%) of embryogenic callus was induced on Murashige & Skoog (MS) medium supplemented with 0.5 mg/L 2,4-dichlorophenoxyacetic acid (2,4-D) and 0.5 mg/L kinetin (Kn) from in vivo leaf explants. Globular somatic embryos were induced and further matured from such embryogenic calli by subsequent culture on the same medium. The highest number of somatic embryos (48.83 ± 4.6) was recovered from embryogenic calli derived from leaf explants after 6 weeks of culture. Synthetic seeds were produced by encapsulating of torpedo stage embryos in sodium alginate (4% W/V) gel, dropped into 100 mM calcium chloride (CaCl2 · 2H2O) solution. The synthetic seeds were germinated on MS medium. The highest frequency of synthetic seed germination (84%) was observed on MS medium supplemented with 1.0 mg/L BA and 0.5 mg/L NAA. Regenerants were successfully acclimatized under ex vitro condition. This is the first report on synthetic seed production of S. chirayita. Application of these protocols would be helpful in reducing stress in natural habitat, and in long-term storage of elite genotypes through synthetic seed production.

Identification of haploid maize by flow cytometry, morphological and molecular markers

The development of homozygous breeding lines in maize may be accelerated through the use of haploids. Thus, the obtaining and prior identification of haploids generated by the haploid inducer lines is an important factor. The purpose of this study was to identify haploids by flow cytometry and to correlate the nuclear DNA content to the morphological and morphometric traits of the seeds that gave rise to them. In addition, molecular markers were used to confirm the androgenetic nature of the haploid. The seeds obtained were derived from the cross between the inbred line W23 and the commercial hybrid P30F90. Among these seeds, a group was selected, putative haploids, whose embryo was white and the pericarp purplish. This group, consisting of 330 seeds, was characterized based on seed morphology, seed morphometry and nuclear DNA content. Flow cytometry analyses identified four haploids, and all of them were small size plants and had brittle leaves. The weight, length, thickness and width of the haploid seeds were very variable indicating that morphometric traits do not constitute reliable data for visual selection of haploid seeds. Based on results, the inbred line W23 induced haploid maize even under tropical conditions. Microsatellite molecular markers (SSR) proved to be efficient, confirming the androgenetic trait of the haploids.

ABSTRAKKomposisi media dasar sangat menentukan keberhasilan proses regenerasi di dalam kultur in vitro. Tujuan penelitian adalah mengetahui pengaruh komposisi media dasar pada media induksi kalus primer dan jenis eksplan terhadap pembentukan embrio somatik kakao. Penelitian dilaksanakan di Laboratorium Kultur Jaringan Unit Pengembangan Benih Unggul Pertanian Balitbangtan, Bogor, mulai bulan Juni 2014 sampai Desember 2015. Induksi kalus primer dari eksplan mahkota bunga dan staminoid kakao klon ICCRI 4 menggunakan dua jenis media dasar, yaitu media Driver dan Kuniyuki (DKW) + 2,4-D 9 μM + kinetin 1,16 μM dan woody plant media (WPM) + 2,4-D 9 μM + kinetin 1,16 μM. Setelah 14 hari, kalus disubkultur pada media induksi kalus sekunder (WPM + 2,4-D 9 μM + kinetin 0,58 μM), kemudian pada media DKW tanpa zat pengatur tumbuh untuk menginduksi pembentukan embrio somatik. Penelitian disusun dalam rancangan faktorial dua faktor dengan 5 ulangan. Faktor pertama adalah jenis media dasar pada media induksi kalus primer (DKW dan WPM), sedangkan faktor kedua adalah jenis eksplan (mahkota bunga dan staminoid). Hasil penelitian menunjukkan terdapat pengaruh interaksi yang nyata antara jenis media dasar dan jenis eksplan terhadap pembentukan kalus dan embrio somatik kakao. Persentase pembentukan kalus paling tinggi diperoleh pada penggunaan media dasar DKW dengan eksplan staminoid (92,5%). Akan tetapi, persentase pembentukan embrio somatik dan jumlah embrio somatik per eksplan paling tinggi diperoleh pada perlakuan media dasar DKW dengan eksplan mahkota bunga (36,5% dan 2,30). Dengan demikian, penggunaan media dasar DKW dan eksplan mahkota bunga lebih direkomendasikan untuk menginduksi pembentukan embrio somatik kakao klon ICCRI 4.
